might be a dead plug?
do a compression test on that cylinder if its not a dead plug. plugs can show spark just slipped in the lead but can fail under compression.
is it back firing or coughing through the carby,
backfire = leaking exhurst valve.

if you put your hand flat over the tip of exhurst pipe (when cold) does it suck your hand onto it every 5 fires???

cough through carby = leaking inlet valve.
if its a leaking valve its possible to lap a valve without taking the head off,make sure the piston in that cyl is up top,  just put a fitting into that plug hole with 150psi pumping continuous into the cylinder,  remove the valve cletes and spring etc.
u can make a fitting by smashing the white porcilin off an old plug and welding a compressor fitting onto it.

take all precautins not to drop that valve into the cylinder, put a few laps of masking tape around the valve stem above the valve stem guide to hold the valve up.

u got 1 number 2 many in brother steve,s number 2,  ;)

just a few things i thought of instantly through same experiances

cheers marty

Yeah could be a plug, swap it with another one and see if the symptoms move following the plug.

Then plug leads, one could be faulty.

Or points worn abit.

If none of that - compression test next, could be a burnt or sticking valve

yep could be cap,
caps can get hairline  cracks and fill up with lead dust from the coil button and redirect the spark somewhere u dont want it to go.
thus not firing right on that cyl.
